Isolation and Identification of 8-glucan Degrading Enzyme Producing Bacterium Using coloured 8-glucan



Abstract

A bacterium K-4-3, producing glucan hydrolyzing eaayme, was isolated from soil and identified to be Bacillus subtilic by its morphological and physiological characteristics. P-glucan was coloured using cibacroa blue 3G-A and cross linked by the addition of 1,4-butaoedioldiglyddyl ether. This substrate was used for the Isolation of 8-glncaaase producing microorganism. The fl-glucan hydrolyzing enzyme activity from Isolated K-43 strain was also measured using the modified substrate. Bacillus subtilis K-4-3 produced the highest extracellalar fl-glucan hydrolyzing activity in the basal medium containing f glucan as a carbon source, peptone and tryptone as a nitrogen source, and magnesium sulfate as an inorganic salt. The optimum temperature and initial pH for f-glucauase production by Bacillus subtilis K-4-3 were 37¡ÆC and pH6. The highest enzyme activity was obtained at the culture age of 54 brs with rotary shaking at 37¡ÆC. The crude enzyme showed the highest activity at pH 7.5-3.0 and 65 ¡ÆC.
